Standards for Histocompatibility Testing Section G - Mixed Leukocyte Culture Tests G1.000
Mixed Leukocyte Culture (MLC) Test G1.100
At the start of culture, lymphocyte viability should exceed 80%. G1.200
Serum used in the culture medium must be screened for support of
cellular proliferation and the absence of cytotoxic antibodies and
must be sterile. G1.300
MLC cultures must be incubated for the length of time shown to give
appropriate proliferation. G1.400
The negative control for each responder cell must consist of responder
cells stimulated with autologous cells. G1.500
The positive control for each responder cell must consist of either of
the following: a) responder cells stimulated with cells from three or
more unrelated individuals; or, b) responder cells stimulated with
cells from two unrelated individuals and a pool of cells from at least
three other individuals. G1.600
In each MLC test, stimulator cells must be shown to be capable of
stimulating unrelated cells. G1.700 An MLC evaluating the potential for rejection is invalid if the cells from the potential recipient do not respond to unrelated stimulator cells. An MLC evaluating the potential for graft versus host disease is invalid if cells from a potential recipient fail to stimulate unrelated cells. |
