BEADCHIP PANELS FOR HIGH THROUGHPUT DONOR MOLECULAR TYPING.

We have developed and tested BeadChips™ comprising locus-specific panels for high throughput low-intermediate resolution donor molecular typing of human leukocyte antigens using BioArray Solutions’ random encoded array format. The HLA- A, B and DR-B panels each are composed of an array of at least 60 encoded types of microparticles ("beads") which display sequence-specific capture probes for multiplexed hybridization-mediated analysis. Using BioArray Solutions’ Thermal BeadChip™ Incubator and automated Array Imaging System and automated allele assignment, 196 samples are readily typed within 8 hours. Parenthetically, single samples can be assayed in less than two hours. The validation of the new BeadChip panels in comparison to conventional SSOP protocols on hundreds of clinical samples show high concordance. High resolution assays also are described which combine hybridization-mediated multiplexed analysis of polymorphisms (hMAP) with on-chip enzyme-mediated analysis of polymorphisms giving superior resolution of typing compared to current methods. BeadChips give rapid, accurate, and inexpensive new platform technology for HLA molecular typing .
