HLA CLASS I ALLELES IN LATIN-AMERICAN POPULATION

Thirty-five Latin-American laboratories registered to the 7th Latin-American Workshop, to DNA-typing HLA Class I genes (A, B and C) and estimate the allele and haplotype population frequencies in Latin-American ethnic groups. Each laboratory typed at least 100 samples of healthy-unrelated individuals using PCR/SSOP. Data from 1550 samples (twenty laboratories) were received for analysis. It was possible to detect 42 alleles in A locus, 72 in B locus and 26 in C locus. In Caucasian populations (Argentina and Brazil) the most frequent alleles were: HLA-A*0101, 0102, 0214, 0301, 2401, 3112; HLA-B*0702, 0801, 13, 14, 1503, 3503, 4402; HLA-C*01, 0101, 0202, 0303, 0702. In Mestizo populations (Mexico, Brazil, Venezuela, Colombia): HLA-A*0101, 3001, 2401, 26, 0201, 29; HLA-B*1503, 1520, 1522, 2702, 3503, 3512; HLA-C *0302, 04, 07, 1204, 0403, 0601, 0801. In mulatto population from Colombia: HLA-A*0202, 2301, 2407, 6601, 3601, 3112; HLA-B*1503, 0702, 3503, 4402, 5301; HLA-C*0302, 0403, 0501, 0704, 0802, and 1601. The haplotypes with frequency > 2% were: HLA-A*0101-B*0801-C*07; HLA-A*02-B*4402-C*07; HLA-A*0301-B*0702-C*0702; HLA-A*3112-B*0702-C*0702 in Caucasians. HLA-A*24-B*0702-C*0703; HLA-A*02-B*1303-C*1204; HLA-A*02-C*0403; HLA-A*3001-C*04; HLA-A*24-B*0702-C*0702 in Mestizos. Haplotypes HLA-A26-B*3801-C*0501; HLA-A*68-B*4002-C*0302 and HLA-A*0202-B*4402 were detected in mulattos. Haplotype analysis suggested that the Caucasian component is greater in populations from Argentina (60%). The Mongoloid component reach 50% in mexican Mestizos. As expected, African component in Colombian mulattos was more evident than that observed in other Mestizo groups. The technique tested in this workshop could be useful for routine typing of HLA-A, B and C alleles, although some samples may require better resolution.
